چکیده

We report the complete genomic sequence of the murine haemopoietic stem cell inhibitor gene (SCI) (1). The SCI protein is identical to the previously identified macrophage inflammatory protein la (MlP-la) (1,2). Using a MIP-la cDNA clone (2), we isolated a single clone from a murine (DBA/2J) adult liver genomic library (Clonetech Labs., USA) which contains the complete SCI/MIP-la gene. The amino acid sequence encoded is shown, and the complete mRNA sequence shown in bold. The putative TATA box, and the (TATTTATT)n repeats in the 3' untranslated region of the mRNA sequence which have been implicated in cytokine mRNA stability (2, 3), are underlined. ACKNOWLEDGEMENTS

STAT3 as a Key Factor in Tumor Microenvironment and Cancer Stem Cell

Background Recent studies revealed that tumor-associated macrophages (TAMs) play a decisive role in the regulation of tumor progression by manipulating tumor oncogenesis, angiogenesis and immune functions within tumor microenvironments. Signal transducer and activator of transcription 3 (STAT3), which is a point of convergence for numerous oncogenic signalling pathways, is constitutively activ...

Transforming growth factor beta: is it a downregulator of stem cell inhibition by macrophage inflammatory protein 1 alpha?

Transforming growth factor beta 1 (TGF-beta 1) and macrophage inflammatory protein 1 alpha (MIP-1 alpha) have recently been identified as potent inhibitors of hemopoietic stem cell proliferation. From previous studies, these molecules appear to have similar functions in the control of stem cell proliferation. This study was designed to investigate the relationship, if any, between these two neg...

Demonstration of stem cell inhibition and myeloprotective effects of SCI/rhMIP1 alpha in vivo.

The proliferative status of the stem cell compartment is thought to be controlled by both positive and negative regulators of proliferation. These regulators have obvious clinical potential in manipulating the integrity and proliferative status of the stem cell in vivo during patient treatment for neoplastic disease. We have tested the ability of the human recombinant homologue of murine macrop...

Maintenance of murine long-term repopulating stem cells in ex vivo culture is affected by modulation of transforming growth factor-beta but not macrophage inflammatory protein-1 alpha activities.

Transforming growth factor-beta (TGF-beta) and macrophage inflammatory protein-l alpha (MIP-1 alpha) are both well-described inhibitors of committed and multipotential hematopoietic progenitors. The effect of these cytokines; on true stem cell activity in ex vivo culture systems as assayed by murine long-term repopulating activity (LTRA) has not been examined. We studied the stem cell effects o...

RAPID COMMUNICATION BB-10010: An Active Variant of Human Macrophage Inflammatory Protein-la With Improved Pharmaceutical Properties

The stem cell inhibitor, macrophage inflammatory proteinla (MIP-la) or LD78, protects multipotent hematopoietic progenitors in murine models from the cytotoxic effects of chemotherapy. Clinical use of human MIP-la during chemotherapy could therefore lead to faster hematologic recovery and may allow dose intensification. We have also shown that human MIP-la causes the rapid mobilization of hemat...
